Comments: | Restriction digests of the clone give the following sizes (kb): BglII--7.2, 2.9; EcoRI--10.0, HindIII--5.2, 2.9, 1.8; XhoI--10.0; PstI--3.8, 2.4, 1.7, 1.3, 0.8. The insert contains the following restriction sites (kb from the EcoRI end): PvuII--0.617, 1.838, 2.447; BglII--0.693, 0.719, 0.801; KpnI--1.324, 3.248; PstI--1.613, 1.833, 2.617, 3.497. The KpnI/PstI fragment (probe A1, nt 3249-3497) and the PvuII/KpnI fragment (probe A2, nt 2448-3248) cross-hybridize to hamster DNA and detect a number of variable fragments in human DNA. A2 contains a CpG island. The PvuII fragment (probe B, nt 1838-2448) does not cross-hybridize to hamster DNA. The EcoRI/BglII fragment (probe C, nt 1-693) does not cross-hybridize to hamster DNA. It detects a 6.0 kb poly(A)+ RNA in HeLa cells. The Alu repeat extends from nt 1368 to 1695. Generated from yHT1 (a 5.5 kb EcoRI/telomeric fragment ligated to the EcoRI/BamHI fragment of pYAC4) by digesting the linear DNA with Bal31 and self-ligating. There are residual yeast/Tetrahymena telomeric repeats located next to the vector HindIII site which is 0.9 kb from the PstI site at nt 3499 of the insert. |
