| Comments： | Restriction digests of the clone give the following sizes (kb): EcoRI--3.7, 0.7; HindIII--4.4; PstI--4.4; BamHI--4.4; EcoRI/HindIII--2.7, 1.0, 0.7. The invariant fragments are derived from the acrocentric chromosomes (13, 14, 21, and 22). The entire plasmid was labeled by the random primer method. Repetitive sequences were blocked by preannealing the probe with human DNA. Hybridization was at 65C for 14 hr. Blot were washed 3 times in 0.5 X SSC (0.1% SDS, 65C). Enzyme(s) not detecting polymorphism: PstI. |
