Comments: | Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--3.0 (doublet); BamHI--3.0, 2.0, 1.0; SalI--6.0; XhoI/PstI--3.0, 1.8, 0.55 (doublet); XbaI--3.0 (doublet). This was constructed from 3 overlapping cDNAs. When transcribed and translated in vitro, a polypeptide of 80 kDa recognized by anti-PFKL antibodies is generated. The clone detects the following genomic restriction fragments (kb), with those not from PFKL indicated by a *: BamHI--8.6, 7.5, 5.0, 4.2*, 3.9*, 3.3, 2.9, 2.2*; KpnI--14, 11, 4.2, 3.3*, 2.6; HindIII-- >20, 12.3, 5.2. A fragment specific to PFKL can be generated from 3 sequences. The insert contains the complete coding sequences, with 5 and 3 untranslated sequences. The KpnI polymorphic site is located in the 3.3 kb BamHI genomic fragment . |
