Comments: | Restriction digests of the clone give the following sizes (kb): EcoRI--19.0, 8.0, 5.5, 4.5, 2.5, 2.4, 2.2, 1.9; BamHI--17.0, 11.0, 7.0, 6.4, 3.1, 1.9; HincII--8.4 (doublet), 6.5, 3.3 (doublet), 3.0, 1.8, 1.75, 1.65, 1.4, 1.3; HindIII--11.0, 6.5, 6.0, 5.4, 4.0 (doublet), 3.5, 2.1, 1.8; BglII--17.0 (doublet), 3.0 (doublet), 2.6. The insert can be excised from the vector with EcoRI, resulting in a 5.5 kb EcoRI vector fragment. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture.
