Comments: | The following enzymes cut the insert once (nt from 5 end, short leader): BssHII-139 MluI-147 XmaIII-167 BglII-228 ScaI-443 Eco47III-470 XbaI-562 NcoI-635 BclI-687 SstII-714 SplI-840 Asp718-1209 KpnI-1213 ApaLI-1996 NsiI-2246. Restriction digests of the clone give the following sizes (kb): EcoRI--2.9, 2.6; SacI--3.5, 1.5, 0.45; EcoRI/SacI/XhoI--2.9, 1.5, 0.7, 0.45; BglI--4.3, 1.2. The insert includes the entire coding sequence of the gene. There are two alternate cDNAs for F7, one encoding a leader of 60 amino acids and one a leader of 38 amino acids. This clone encodes the short leader. The depositor indicates the polymorphism has allele lengths that differ by only a few nucleotides. It would be difficult to detect this polymorphism by means of a Southern blot of the genomic digest. |
