Comments: | Insert contains AvaI (2), PstI, KpnI (2), and BamHI sites. Restriction digests of the clone give the following sizes (kb): EcoRI--5.6; PstI--3.0, 2.6; BamHI--3.9, 1.7; HindIII--3.5, 1.95; AvaI--2.7, 1.5, 1.2. Detects loss of heterozygosity in renal cell carcinoma. Derived from Ch4A-lambdaH3 (DNF15S2, formerly D1S1, p01970). This probe detects a genomic HindIII fragment of 2 kb from chromosome 3 and genomic HindIII fragments of 3.8 and 8 kb from chromosome 1. The copy number of the fragments on chromosome 1 is 6-8 per haploid genome. This probe detects a StuI RFLP mapping to 1p36 and a HindIII RFLP mapping to 3p21. Can detect loss of heterozygosity in lung cancers. The StuI polymorphism shows autosomal dominant inheritance because of the repetitive nature of the locus. Band sizes are 3.8 kb, 3.6 kb, 2.0 kb, and 1.2 + 0.6 kb. |
