Comments: | Restriction digests of the clone give the following sizes (kb in order from the CEN end): NruI--100, 130, 40, 410; MluI--230, 105, 100, 245. A complete cDNA probe detects a 70 kb BssHII fragment in both YAC and human DNA. The gene is located near the center of the insert, with the 5 end toward the yeast centromere. It is contained in a 100 kb MluI fragment. When transferred to mouse L-cell line A-9 (GM00346B, HPRT-negative), the YAC conferred the ability to grow in HAT medium. |
