| Comments： | Restriction digests of the clone give the following sizes (kb): PvuII--2.5, 1.5, 1.3; PstI--4.1, 1.3; KpnI--4.1, 1.2; EcoRI/XhoI--3.0, 2.3; BglI--3.7, 1.2, 0.4. Transcription from the T3 promoter gives the sense strand. The plasmid can be linearized with XhoI. The insert contains the following restriction sites (approximate kb from the 5 end): BglI--2.16; EcoRV--2.29; KpnI--1.21; NcoI--2.09, 2.18; PstI--1.30; PvuII--1.05; SacI--0.01. The protein differs from the human GRB2 sequence only at aa 154, where the murine protein has Met instead of Val. Detects transcripts of 3.8 kb and 1.5 kb in the following mouse tissues: lung, heart, testis, muscle, cerebellum, brain, kidney, spleen, ovary, thymus and at low levels in the liver. Constructed by ligation of a 5 155 bp EcoRI-AflII fragment, amplified from a kidney cDNA library, and a 3 2,206 bp AflII-XhoI fragment, isolated from a brain cDNA library. |
