Comments: | Restriction digests of the clone give the following sizes (kb): BamHI--7.6, EcoRI--2.2 (doublet), 1.7, 1.4; KpnI--7.6. The URA3 marker is from S. cerevisiae but complements the ura4 allele from Sc. pombe. Expression of chloramphenicol acetyltransferase is hormone-inducible in both S. cerevisiae and Sc. pombe hosts, in the presence of a glucocorticoid receptor such as that encoded by pG-N795 (ATCC 37813) or pART1/N795 (ATCC 37814). Contains the CYC1 TATA region from S. cerevisiae (nt -178 to +49) fused at nt -178 to 3 copies of a 26 bp glucocorticoid response element (GRE) from the rat tyrosine aminotransferase gene. A BglII/BamHI fragment with a chloramphenicol acetyltransferase coding region, its own AUG codon, and some SV40 sequences was cloned into the BamHI site of the polylinker. The other polylinker sites are 3 to this insert. |
