Vector Information: | Size (kb): 4.5000000000000000 Vector: pUB110cop1 (plasmid) Promoters: Promoter none Construction: pUB110, pEB114cop1 Marker(s):kanR,neoR Construct size (kb): 4.5 Features: marker(s): kanR, neoR promoter: none replicon: pUB110 |
Applications: | shuttle vector vector with low copy number |
Comments: | Restriction digests of the clone give the following sizes (kb): EcoRI--4.5; NcoI--4.5; BamHI--4.5. The copy number is 11 +/- 2 in Bacillus subtilis, resulting in an increase in structural and segregational stability. Low copy number shuttle vector. Constructed by replacing the NcoI/AflII fragment of pUB110 (nt 4033-4543) with the corresponding fragment from pEB114cop1 which contains a G -> T transversion in the Shine-Dalgarno sequence of repU at position 4130. |
