Comments: | Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--4.4, 2.6; EcoRI--7.0; HindIII--7.0; BamHI--7.0; PstI--6.6, 0.4. The insert is the 4.2 kb HindIII/EcoRI fragment containing VTR B and exons 4 and 5. The repeat elements are in the HindIII/XhoI fragment of about 1.8 kb. The exons are close to the EcoRI end, with exon 5 contained in a SstI/EcoRI fragment of about 0.3 kb. BamHI, BglI, BstEII, EcoRI, HindIII and SstI cut outside the VNTR, generating one fragment per allele, but the sizes are difficult to resolve. A 0.6 kb TaqI fragment has been used to detect the VNTR polymorphism. TaqI cuts within the VNTR generating 1 to 4 different sized alleles per individual. HinfI also cuts within the VNTR. |
