Comments: | The insert contains the following restriction sites (in approximate kb from the end upstream of the gene): EcoRI--2.5, HpaI--4.0; PstI--6.7; BglII--7.3; HindIII--8.0; EcoRI--10.0. Restriction digests of the clone give the following sizes (kb): EcoRI--23.0, 9.0, 3.5, 3.0, 2.3; BamHI-->23.0, 11.0; HindIII-->23.0, 9.0, 4.8, 3.1. The central EcoRI fragment of this construct, and a HpaI/EcoRI fragment derived from that, contain the sequences encoding the complete polypeptide. The insert contains 2 open reading frames. Sequences up to a BalI site at 5.5 kb from the upstream region and after a StuI site in the second reading frame can be deleted without destroying the toxicity of the product to mosquito larvae. |
