Recommendations for Infection: Plant cells 24-48 hours in advance and infect when cultures are 60-90% confluent. Remove medium and inoculate with a small volume (e.g. 1mL per 25 cm squared) of virus diluted with VGM to provide a MOI of about 0.1-1.0. Adsorb 1-2 hours at 37°C in a 5% CO2 atmosphere. End adsorbtion by adding VGM.
Incubation: Incubate infected culture for 1-6 days at 37°C in a 5% CO2 humidified atmosphere until CPE is well advanced through 90% of culture. |
