| Comments: | Restriction digests of the clone give the following sizes (kb): EcoRI--3.0; BamHI--3.0; EcoRI/BamHI--2.8, 0.20; PstI--3.0; HindIII--3.0. The pseudoautosomal boundary lies proximal to the sequences defined by pHf0.2 between the EcoRI and RsaI sites. In RsaI digests, detects 2.4 and 2.8 kb bands in males, but only 2.8 kb band in females (2.4 kb band is Y-specific, 2.8 kb band is X-specific). DNA from male and female humans, gorillas and chimpanzees show fragments of identical sizes. Hybridizes to 1.8 kb band in EcoRI digests, and 0.6 kb in HaeIII digests for both males and females. Detects 3.3 and 4.5 kb bands in male SacI but only 4.5 kb in female SacI digests (3.3 kb band is Y-specific, 4.5 kb is X-specific). Sequence analysis indicates a small amount of Alu repeat sequence on the probe. The polymorphic MspI site detected by this probe lies between the probe sequences and the pseudoautosomal boundary, 181 bp from a non-polymorphic MspI site in the pseudoautosomal region. |
