| Comments: | Restriction digests of the clone give the following sizes (kb): PvuII--2.5, 0.5, 0.3; PvuI--2.2, 1.2; PvuI/EcoRI--1.7, 1.1, 0.6; PstI--3.4; PvuII/XbaI--2.5, 0.3, 0.3, 0.2. Detects sequences from the Op (between O12 and O13) and Od (between O2 and O2) regions. Does not cross-hybridize to the W region. The EcoRI polymorphism can be used only for haplotype G and the BglII polymorphism can be used only for haplotype N. In the KpnI/BglII and KpnI/SacI digests, the KpnI site is polymorphic. Contains the SalI fragment from m144-2 which had been generated by adding SalI linkers to an RsaI fragment. During recloning, the SalI end 150 bp from the vector PvuII site was destroyed. Neither KpnI, ApaI nor XhoI cut at this position. The insert is not readily excised, but can be amplified with standard M13 sequencing (forward and reverse) primers. |
