TaqI and MspI RFLP alleles are in strong linkage disequilibrium. Codominant segregation was observed in 4 unrelated families (49 individuals). The insert contains ATG at 97, TGA at 610, and 89 bp of 3 untranslated sequence including AATAA followed by poly(A) 17 bp downstream. Recorded as not polymorphic by Collaborative Research, meaning either not polymorphic in 0.8% agarose gels, or difficult to work with. Plasmid pPGBC9 was cut with BamHI and the fragments of 0.5 kb and 0.6 kb were isolated, pooled and used as the probe for the MspI and TaqI RFLPs. Enzyme(s) not detecting polymorphism: RsaI, BstEII, EcoRV, BamHI, HindIII, BglII, PvuII, BglI and PstI in at least 6 unrelated individuals. |
