| Comments： | Host containing defective prophage for use with vectors regulating expression from the lambda PL promoter. Encodes cI857 but not a functional N gene. Phenotype verified: galK(am), strR, kanR, thi, leu. Because of the temperature sensitive repressor, heat shock for CaCl2 transformations should be at 32C for 10 min, followed by 1 min on ice. Add LB and grow at room temperature for 1-2 hours. Selection for ampR is on LB + amp (100 ug/ml) plates incubated at 30-32C. An isogenic host with N gene function is TAP56, ATCC 47074. |
