To use as a template, the plasmid must be linearized with either PstI or HindIII. A partial length (about 260 bp) fragment of Hop Stunt Viroid cDNA was cloned into BamHI/PstI of pSP65; noninfectious. Constructed by R.W. Hammond. E. Shikata provided the original Hop Stunt Viroid cDNA. |
