| Comments： | Restriction digests of the clone give the following sizes (kb): EcoRI--10.0, 7.6, 6.5, 5.1, 3.65, 2.4; SalI--30.0+; BamHI--25.0, 3.3, 2.45, 2.75, 2.4; XbaI--18.5, 8.7, 4.0, 2.4, 1.85. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture. Codominant segregation demonstrated in 48 3-generation families. cEFD52 was pre-associated with excess human DNA before hybridization. Otherwise, RFLPs were observed under normal conditions of hybridization and washing. |
