| Comments: | Among the restriction sites in the insert are the following (in bp from the T7 promoter side 5): Pst--317; BglII--321;, NdeI--1273; BglII--1963; PstI--2905. Restriction digests of the clone give the following sizes (kb): EcoRI--3.2, 3.0; HindIII--6.2; PstI--3.2, 2.7, 0.35; BglII--4.7, 1.7; BamHI--6.2. zif268 is induced in mouse 3T3 cells by serum, tetradecanoylphorbol acetate, PDGF, and FGF. Induction is coordinate with c-fos. Genomic 5 flanking regions contain multiple serum response elements (SRE) which compete with the c-fos SRE. Encodes a protein of 533 amino acids with three typical zinc finger sequences. Between the polyadenylation signals at the 3 end of the message are two ATTTA sequences, related to the message instability characteristics of immediate early mRNAs. Homologous to NGFI-A, which is activated in rat pheochromocytoma cells by nerve growth factor. Within the zinc finger region it is related to the Krox gene family (Krox24). |
