For growth of the host strain, the medium must contain DAP (diaminopimelic acid) (100 mg/L) and thymidine (20 mg/L). The insert contains sequences encoding all 191 amino acids of human growth hormone. Contains an internal PstI, BglII and PvuII site. The vector (ATCC 31539, 40012) is pBR322 modified by the insertion of 2 lac UV5 promoters at the EcoRI site. These promoters regulate expression of the insert. |
