| Storage Conditions: | xa0-70°C or colder |
| Comments: | Virus growth is optimal at pH 6.8-7.3, in roller or rocker cultures incubated at approximately 33°C. All members of the rhinovirus group are ether resistant. |
| Effect on Host: | Cytopathic effects (CPE) in culture include rounding and sloughing of cells xa0CPE in culture include cell rounding and sloughing |
| Growth Medium: | ATCC® Medium 30-2003: Eagles Minimum Essential Medium (EMEM) Fetal Bovine Serum |
| Recommended Host: | Production Host - H1-HeLa cells (ATCC CRL-1958)
Recommended Host: xa0H1 HeLa (ATCC® CRL-1958x99) Alternate Host(s) - WI-38 cells (ATCC CCL-75); MRC-5 cells (ATCC CCL-171); HeLa cells (ATCC CCL-2); other human cells |
| Growth Conditions: | Host - H1 HeLa cells (ATCC CRL-1958)
Medium - EMEM (ATCC 30-2003) + 2% FBS (ATCC 30-2020) Infection conditions - For best results, plate cells 24-48 hours in advance. Infect at about 80% confluence using a multiplicity of infection (MOI) of 0.1 to 1.0. Adsorb virus to cells in a small volume for one hour. After adsorbtion, feed with sufficient medium to maintain cells and incubate for 1 to 7 days at 33 °C, with rocking, in a humidified atmosphere with 5% CO2 . Harvest culture fluid or fluid and cells when entire culture displays cytopathic effects. Preservation - Freeze as quickly as possible and store at -70°C or colder. |
