| Comments: | Restriction digests of the clone give the following sizes (kb): SacI--3.0, 1.35; SacI/PstI--3.0, 0.9, 0.4; PstI--3.8, 0.6; EcoRI--3.36, 0.38 (doublet), 0.3; HindIII--4.3. There is tight linkage between multiple endocrine neoplasia type 2A (MEN2A) and D10Z1 (theta = 0, LOD score = 12.02). Data for haplotypes formed by using the A and F morphs improved the LOD score for linkage between multiple endocrine neoplasia type 2A (MEN2A) and D10Z1 (theta = 0, LOD score = 14.06). There is tight linkage between medullary thyroid carcinoma (MTC) with parathyroid tumors (PTs) and no pheochromocytomas (PHEOs) and D10Z1 (theta = 0, LOD score = 2.86). The insert contains 4 EcoRI repeat units of 340 bp each. A SacI/PstI double digest should give fragments of 1.0 kb and 0.36 kb from the insert. High stringency hybridization and washing conditions are necessary due to the repetitive nature of the probe. The HinfI polymorphism detects the haplotypes at the following frequencies: F+G+ (0.5), F+G- (not seen), F-G+ (0.02), and F-G- (0.48). |
