An artificial polyA tail was added onto the 3 end of the insert and can be excised using BamHI + NotI. XhoI + BamHI will excise insert without the polyA tail. The insert contains the following restriction sites (approximate kb from the 5 end): HindIII--0.16; PstI--0.18; ClaI--0.47; BglI--0.54. Restriction digests of the clone give the following sizes (kb): NotI--4.0; NotI/XhoI--3.0, 1.1; XhoI--4.0. |
