| Comments： | Restriction digests of the clone give the following sizes (kb): BamHI--5.6, 0.3; EcoRI--6.2. This is a promoter-cloning vector with beta-galactosidase reporter gene, trp a terminator upstream of MCS, galK leader with stop codons in 3 reading frames, and intergenic region of M13 to permit ssDNA preparation for mutagenesis and/or sequencing. RL724 (TS164827) and RL726 (ATCC 37684) differ in having the M13 intergenic region in opposite orientations in a different sized fragment. This plasmid was constructed from the galK leader of pKO100 fused to the lacZ of pMC1403; the M13 intergenic region of pZ152; the multiple cloning sites of M13mp19; and the trp a terminator of pRL118. |
