| Growth Conditions: | Temperature: 25.0°C Protocol: This strain is shipped as a frozen stabilate on dry ice. Please read the instructions that accompany the shipment before handling the material. Thaw the vial as instructed and transfer its entire contents to 10 ml of ATCC medium 1967 in a T-25 tissue culture flask. The bacterial flora present in the shipped culture will support growth, but growth may be improved by using medium that has been bacterized with Enterobacter aerogenes ATCC 13048 approximately 24 hours before inoculation with the protist. (Other species of bacteria may work equally well, but this one has been empirically assessed.) Subculture every 14-21 days. To subculture, aseptically scrape the bottom of the flask, agitate the flask to distribute the cells evenly, and aseptically transfer 0.25 ml to 10 ml of bacterized medium in a T-25 flask. Screw the cap on tightly and incubate the flask at 25C. |
