| Comments: | Restriction digests of the clone give the following sizes (kb): EcoRI--4.3, 1.7; HindIII--5.6, 0.5; BamHI--4.3, 1.2, 0.3; PstI--6.0. The insert contains the following sites (nucleotide position according to the published sequence): BamHI--514, 848; HindIII--1240, XbaI--1480. Crry and Cr-2 are within 10 kb of each other and are transcribed in the same direction. Encodes a single open reading frame starting at nt 482 of the published sequence and extending through the 3 end of the cDNA. Comparison with genomic sequence shows this clone lacks sequences for 7 additional amino acids. Spleen 5 cDNA (ATCC 63065) differs from liver 10 cDNA (TS168429) by having an additional 129 nucleotides between the signal sequence and the nucleotides encoding the first 60 aa repeat. Spleen 5 also does not extend as far in the 5 direction. Liver 10 cDNA and spleen 5 cDNA are clones of alternative splice products of the Crry gene. The mRNAs are transcribed in all tissues examined in a 10 (liver 10) to 1 (spleen 5) ratio. When the 340 bp BamHI/BamHI fragment (lacking the extra 129 nucleotides) is used as a probe, the following EcoRI fragments are detected in BALB/c mice: 7.5 kb, 5.6 kb (chromosome 8), and 4.5 kb. Detects mRNA species of 1.9, 2.0, and 2.1 kb. |
