| Comments： | Restriction digests of the clone give the following sizes (kb): EcoRI--6.6, 3.5; HindIII--6.0, 1.9, 1.4, 0.8; PstI--6.3, 3.8; KpnI--10.1. The insert contains the following restriction sites (approximate kb from the end upstream of ccrA): PstI--0.5, MboI--0.9, 1.9; KpnI--1.2; HindIII--3.6, 5.4. The 941 bp MboI fragment contains the complete gene, with 150 bp 5 and 50 bp 3 flanking sequence. The sequence of most of this fragment has been reported. The vector is a derivative of pMK16 deleted for the carboxy terminal half of the tetR marker and has had the small EcoRI/BamHI fragment replaced with the MCS-containing EcoRI/BamHI fragment from plink 322. |
