The insert contains the following restriction sites (approximate kb from one end): HindIII--0.3 (spleen DNA), 7.8, 9.3 (viral); KpnI--2.7, 3.6, 5.6 (viral), 14.5 (spleen); SalI--3.0 (viral); XhoI--6.7 (viral); SacI--2.0, 2.8, 9.9 (viral), 11.3, 13.2. Size of construct: 19.7kb. Full-length clone of reticuloendotheliosis virus-A (pSW253) inserted at EcoRI site in pBR322; construct is amp resistant and tet resistant. See also GenBank/EMBL/DDBJ J01749. Corresponds to the larger EcoRI fragment of clone 2-20-6. GenBank K02357 begins 170 bp upstream of the XhoI site. The insert contains the 8.3 kb REV-A proviral DNA and flanking chicken spleen sequences subcloned from clone 2-20-6. The proviral sequences begin about 2 kb from the EcoRI site closer to the HindIII site. |
