| Comments： | Restriction digests of the clone give the following sizes (kb): EcoRI--11.0; HindIII--6.6, 4.1; KpnI/SphI--7.2, 3.6; PstI--7.6, 3.0; XbaI--11.0. The 3.5 kb SphI/KpnI fragment conferring mevinolin resistance has been sequenced. The mutation in the hmgA gene is at position -29 relative to the transcription start site, and results in an increased level of transcription. Shuttle vector with demonstrated ability to transform Haloferax volcanii, Haloferax mediterranei (ATCC 33500), Halobacterium halobium, Haloarcula hispanica (ATCC 33960), and Haloarcula vallismortis (ATCC 29715). The order of the major features in this plasmid is: NcoI - BamHI - HindIII - XbaI - KpnI - mevR - sphI - pMB1 ori - ampR - EcoRI - ClaI - HindIII - pHV2 fragment. |
