| Comments: | Restriction digests of the clone give the following sizes (kb): XhoI--3.05, 2.55; BamHI--3.2, 1.8, 0.65; KpnI--5.7; HindIII--4.0, 1.7; PvuII--5.7. The non-vector XhoI fragment contains the following restriction sites (approximate kb from the 5 end): BamHI--0.24, 0.84, 2.45; KpnI--0.92; HindIII--1.3; SacI--1.6; PuvII--0.05, 2.24, 2.28. A 1.7 kb BamHI fragment contains the entire coding region. Digestion with XhoI releases a 2.4 kb fragment that contains the insert plus approximately 200 bp of vector. The 0.7 kb SacI/PvuII fragment from within the coding region can be used as a hybridization probe. |
