The insert contains the following restriction sites (approximate kb from the 5 end): BglI--0.18, 0.37; SalI--0.34; SacI--1.11. Digestion with XbaI yields an intact insert with a portion of the T7 phi 10 promoter included at the 5 end and a portion of the MCS at the 3 end. Extraneous 3 material can be removed with an EcoRI digest. The pH 2.5 acid phosphatase is more active as a phytase. |
