| Comments: | Restriction digests of the clone give the following sizes (kb): EcoRI/SalI--2.6, 1.4; EcoRI--4.0; PstI--2.6, 1.4. The cre gene contains the following restriction sites (approximate kb from the 5 end): BamHI--0.84; ClaI--1.29. The lambda/plasmid hybrid vectors can then be used for automatic subcloning; that is, the cre-mediated excision of plasmid constructs. A plasmid useful for generating novel lambda/plasmid hybrid vectors by using cre-lox recombination to exchange new plasmids for plasmids already in lambda/plasmid hybrids. Constructed by ligating a 1.3 kb cre-containing fragment from lambdaKC to pHSG298 digested with EcoRI+SalI. |
