| Comments: | Restriction digests of the clone give the following sizes (kb): HindIII--9.8, 2.8; EcoRI--3.8, 3.2, 3.0, 2.3; PstI--4.8, 3.2, 2.3, 1.8; SalI--13.0; BamHI--4.6, 4.1, 3.8. The insert extends from an EcoRI site to the HindIII site (about 1.5 to 11.5 kb). It includes 3 copies of a 10 bp sequence (-69, -29, +1) found in other ribosomal protein genes. The coding sequence starts in exon 2. Introns 3 and 4 contain Alu-homologous repeats. The 3 flanking sequence contains 10 bp complementary to U1 RNA. A 260 bp AluI/PstI fragment in intron 3 has been shown to be single copy and useful for mapping purposes. It shows only slight cross-reaction with CHO cell (Chinese hamster) DNA. The fragment cloned was an EcoRI/HindIII fragment to which a HindIII linker was added. Thus the clone contains both EcoRI and HindIII sites at its 5 end and a HindIII site at the 3 end. There are several internal EcoRI sites. |
