| Comments: | Restriction digests of the clone give the following sizes (kb): SalI--4.9; BamHI--4.9; EcoRI--4.9; PstI--3.8, 1.1; EcoRI/BamHI--2.7, 2.2. The 850 bp HaeIII fragment is free of repeated sequences. PvuII digests revealed a 5.7 kb fragment with a polymorphic 5.2 kb fragment in 1/33 chromosomes studied. These RFLPs are also seen with human HPRT cDNA but the picture is more complex because of numerous bands. The variant HindIII allele segregates independently of the variant PvuII allele. The 850 bp HaeIII fragment detects the following genomic fragments (kb): EcoRI--8.8; HindIII--9.8; BamHI--3.4 kb. This clone was derived from Hulambda10. Enzyme(s) not detecting polymorphism: TaqI, MspI, PstI, BstNI, XbaI, SacI, HincII, StuII, MboI, EcoRV. |
