| Comments: | Restriction digests of the clone give the following sizes (kb): HindIII--4.4, 3.5; EcoRI--6.1, 1.8; AvaI--4.7, 3.2; BamHI--8.0; PvuII--8.0. The sequences detected by Y-190 are present in numerous XX males. This probe hybridizes to Y-specific HindIII fragments of about 4 and 5 kb (strong) and 9 kb (weak) in the orangutan and 4 kb and 9 kb (weak) in the rhesus monkey. This probe detects 2 blocks of repeats on Yp. The hypervariable major block is composed of tandem 20.3 kb repeats with alleles ranging from about 540 to 800 kb (XbaI). The minor block is not variable and is about 60 kb. Detects a Y-specific 3.5 kb HindIII fragment that has a copy number of about 100 in normal human males. Also detects less repeated HindIII fragments ranging from 1.5 to 5.7 kb. This probe is useful for detection of Y-derived marker chromosomes and mosaicism. |
