| Comments: | The insert contains 1 KpnI site (in the region encoding the triple-helical domain) and 5 PstI sites. Restriction digests of the clone give the following sizes (kb): EcoRI--2.7, 2.0; HindIII--4.7; BamHI/EcoRI--2.7, 2.0; PstI--3.2, 0.8, 0.5, 0.2; XbaI--4.7. COL6A1 and COL6A2 are closely linked (about 200 kb). This probe encodes an epitope of collagen type VI since it was identified by immunoscreening a lambda gt11 library. The insert encompasses that portion of the cDNA from about 1.2 kb to 3.2 kb from the 5 end, including the 3 portion of the triple-helical domain. The probe detects the following genomic TaqI fragments (kb): human specific--6.0, 4.4 and/or 4.1 (polymorphic); mouse--6.6; Chinese hamster--2.0. This probe detects a mRNA of 4.2 kb in human fibroblasts. |
