| Comments： | Restriction digests of the clone give the following sizes (kb): HindIII---4.0, 2.3; EcoRI--6.3; PstI--4.5, 1.8; BamHI--4.7, 1.8; SalI--6.3. This probe codes for 182 Gly-Xaa-Yaa residues, the 229 amino acid COOH-terminal noncollagenous peptide and 435 nucleotides of the 3 untranslated region. This insert includes the 5 portion of the 3 untranslated region and contains only one of the many AATAAA polyadenylation signals of the mRNA. The collagenous region is encoded primarily in the 5 PstI-BamHI (0.52 kb) fragment of the insert. The non-collagenous carboxyl terminus (NC1) is encoded in the adjacent BamHI-HincII fragment (about 0.72 kb). The HincII-PstI fragment contains the 3 untranslated sequences. This probe detects mRNA of 6.5 kb (major) and 5.9 and 5.3 kb (minor) in human skin fibroblasts. |
