| Comments: | Restriction digests of the clone give the following sizes (kb): SalI--23.0, 6.1; EcoRI--22.5, 3.5, 2.9; BamHI--17.0, 6.6, 3.45, 1.95; HindIII--28.8. For expression, foreign DNA should contain the gene of interest as well as a promoter and polyadenylation signals. Maintained as approximately 10 plasmid copies in eukaryotic cells that provide the EBNA-1 gene product in trans. This protein is produced in P3HR-1 cells (ATCC HTB-62). Epstein-Barr virus-based shuttle vector that can be maintained as a plasmid in cells expressing EBNA-1 or induced to replicate lytically by treatment with tetradecanoylphorbol acetate or introduction of a construct expressing BZLF-1. The virus particles can then be used to infect other eukaryotic cells. Encodes all sequences necessary for virus packaging, including the terminal repeats. The order of the major features of the plasmid is: HSV-1 Tk promoter - G418 neo HSV-1 Tk terminators - EcoRI - oriP - EBV genes - oriLyt - SalI - EcoRI - terminal repeats (TR) - ampR. |
