| Comments: | Restriction digests of the clone give the following sizes (kb): AccI--4.6, 1.95; BamHI--2.75, 2.25, 1.35; EcoRI--3.5, 3.3; HindIII--5.1, 1.7; PstI--3.2, 1.2, 0.81, 0.56, 0.36. 0.32. Bands detected after a MspI digest are faint. RsaI digest demonstrates the polymorphism best. The probe should be hybridized to total human DNA (50-100 ug/ml) for 2 h before incubation with the filter (42C) because the probe contains repetitive DNA. Enzyme(s) not detecting polymorphism: BglII, PstI, PvuII. |
