| Comments: | The #4 peak was well-isolated from hamster background with little debris. According to fluorescence in situ hybridization using genomic hamster DNA with sorted chromosomes, there is 17% hamster DNA. About 83% of the library is human (and 17% hamster) by plaque hybridization. Of 2315 randomly selected plaques screened: 224 human single copy greater than 600 bp; 1106-human probably repetitive; 595-inserts absent or less than 600 bp; 62-hamster single copy greater than 600 bp; 328-hamster probably repetitive. There is approximately 20% hamster impurity. Of 47 unique sequence clones tested, 47 map to chromosome 4. The average insert size is 2.0 kb. Of the 224 single copy inserts screened, all detected an appropriate HindIII fragment on chromosome 4; some detected additional fragments not on 4; some also cross-hybridized to hamster sequences. The library LL04NS01, TS192696, 0.8 genomic equivalents, is distributed together with LL04NS02, ATCC 57745, 10.0 genomic equivalents, because there is a possibility that a sequence in one may not be found in the other. |
