| Comments: | Restriction digests on an agarose gel give the following sizes (kb): EcoRI--6.4, 4.1; BamHI--10.5. The sequence of the insert is AATTCATGTT CGTCAATCAG CACCTTTGTG GTTCTCACCT CGTTGAAGCT TTGTACCTTG TTTGCGGTGA ACGTGGTTTC TTCTACACTC CTAAGACTTA ATAGGATC, including a HindIII site at position 46. This construct directs the synthesis of a beta-galactosidase-insulin B chain fusion protein regulated by the lac promoter. The B peptide can be released by cyanogen bromide cleavage and used to reconstitute insulin with the product of pIA1, ATCC 31448. This clone was constructed from pBR322, an EcoRI fragment from lambda plac containing most of the beta-galactosidase gene, and synthetic DNA encoding human insulin B chain. |
