| Comments： | Gene targeting vector for CD40 ligand (CD40L, CD154), useful for preparing CD40L-deficient (knockout) mice. Homologous recombination with the vector results in the removal of exons 3 and 4 from the CD40L gene. The vector can be electroporated into embryonic stem cells, for example at 200 V and 960 uF. Culture the cells on a feeder layer, and select in 175 ug/ml G418+2 mM gancyclovir, for about 10 days. Then plate single clones and analyze via PCR. |
