| Comments： | Restriction digests of pCAS4 give the following sizes (kb): BamHI--10.5; EcoRI--9.0, 1.9; HindIII--9.0, 0.7, 0.7. pLysS is also detected at 5.1 kb with EcoRI or HindIII, or 4.4 and 0.6 with BamHI. Restriction digest of the pCAS4 gives the following sizes (kb): EcoRV--8.2,1.9. The insert differs from that of NEB671 (ATCC 68447) by the modification the sequence encoding aa 141-143 (GATATTGAA) to GCGATCGCA. This changes the aa sequence of the protein from DIE to AIA and inactivates 3 to 5 exonuclease activity. |
