| Comments: | Restriction digests of the clone give the following sizes (kb): EcoRI--5.45, 1.2; HindIII--5.0, 1.65; PstI--6.4, 0.25; BamHI--6.65. EST01828 includes sequences that translate into homeodomains, with the most similarity to Drosophila otd. It also includes a splice acceptor sequence that begins where the sequence of EST1828 diverges from otd. EST01828 matches PIR accession A35912 with 52.77% identity and 75% similarity over the sequence range: 242-135. Isolated from a directional library constructed by B. Soares, Columbia University. First strand synthesis was primed with an oligo(dT)-NotI primer. Much of the priming occurred by the NotI portion of the primer, so although the clones are directional, the NotI site is not necessarily the 3 end of the message. This sequence record is for Drosophila melanogaster orthodenticle (Otd). |
