| Comments: | Restriction digests of the clone give the following sizes (kb): BamHI/BglII--5.0, 0.9; BglII/HindIII--5.3, 0.6; BamHI/EcoRI--4.3, 2.1. The insert contains a HindIII site 0.52 kb downstream of the 5 end BglII site. Transcription from the trp promoter produces a fusion protein including 54 aa from the 5 poliovirus 3C protease (encoded by the vector), 46 aa from the C-terminal portion of gp120, and 240 aa from the N-terminal portion of gp41. The recombinant protein is recognized by monoclonal antibodies specific to gp41 and gp120. The vector is a pBR322 derivative with trp operator/promoter, Shine-Dalgarno and protease 3C sequences inserted at the ClaI site. |
