Derivation:
The WI-38 human diploid cell line was derived by Leonard Hayflick from normal embryonic (3 months gestation) lung tissue.

Comments:
WI-38 cells have a finite lifetime of 50 plus or minus 10 population doublings with a doubling time of 24 hours.

This line was the first human diploid cell line to be used in human vaccine preparation.

The 8th passage ampule from which this freeze was derived was found to contain a bacterial contaminant (a micrococcus). The cell line was subsequently cured by several passages in the presence of antibiotics.

Growth of the cells is enhanced by addition of tumor necrosis factor alpha (TNF alpha) to the medium.

This cell line is negative for reverse transcriptase.

Complete Growth Medium: The base medium for this cell line is ATCC-formulated Eagles Minimum Essential Medium, Catalog No. 30-2003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Subculturing:

Volumes used in this protocol are for 75 cm2 flask; proportionally reduce or increase amount of dissociation medium for culture vessels of other sizes.xa0Corning® T-75 flasks (catalog #430641) are recommended for subculturing this product.

  1. Remove and discard culture medium.
  2. Briefly rinse the cell layer with 0.25% (w/v) Trypsin-0.53mMxa0EDTA solution to remove all traces of serum which contains trypsin inhibitor.
  3. Add 2.0 to 3.0 mL of Trypsin-EDTA solution to flask and observe cells under an inverted microscope until cell layer is dispersed (usually within 5 to 15 minutes). xa0Note: To avoid clumping do not agitate the cells by hitting or xa0xa0shaking the flask while waiting for the cells to detach.xa0 Cells that are difficult to detach may be placed at 37°C to facilitate dispersal.xa0
  4. Add 6.0 to 8.0 mL of complete growth medium and aspirate cells by gently pipetting.xa0
  5. Add appropriate aliquots of cell suspension to new culture vessels
  6. Place culture vessels in incubators at 37°C.
