| Applications: | This construct has been used to immortalize primary epithelial cells of various rat tissues including kidney, liver, heart, pancreas and thyroid. The line was produced by inserting the E1A 12S cDNA sequence into the retrovirus vector DOL. The retroviruses produced during the first 24 hours post-transfection were harvested and used to infect fresh Psi2 cells to generate the Psi2 12S6 line. The retrovirus produced by this line can be used to to immortalize primary epithelial cells of various rat tissues including kidney, liver, heart, pancreas and thyroid. Tested and found negative for ectromelia virus (mousepox). |

| HeLa Markers: | N |
| Genes Expressed: | a retrovirus vector containing the adenovirus 12S E1A and the neomycin phosphotransferase genes,early-region 1A, 12S E1A |
| Cellular Products: | a retrovirus vector containing the adenovirus 12S E1A and the neomycin phosphotransferase genes |
| Comments: | The 13S E1A mRNA encoded by rv13S (ATCC CRL-1809) is 46 amino acids longer than the 12S E1A mRNA encoded by rv12S (TS211080). This construct has been used to immortalize primary epithelial cells of various rat tissues including kidney, liver, heart, pancreas and thyroid. The line was produced by inserting the E1A 12S cDNA sequence into the retrovirus vector DOL. The resulting vector DNA (rv12S) was transfected into Psi2 cells by calcium phosphate precipitation. The insert contains sequences from adenovirus type 2-adenovirus type 5 hybrid cDNAs. |
| Complete Growth Medium: | Dulbeccos modified Eagles medium with 4.5 g/L glucose, 90%; bovine calf serum, 10%
|
| Subculturing: | Subcultivation Ratio: A subcultivation ratio of 1:20 to 1:40 is recommended Medium Renewal: Every 2 to 3 days Remove medium, add fresh 0.25% trypsin solution for several minutes, remove trypsin and allow the cells to sit at 37C until they begin to detach (about 3 to 5 minutes). Add fresh medium, aspirate and dispense into new flasks. |
