| Derivation: | The J.RT3-T3.5 cell line is a derivative mutant of the Jurkat leukemia cell line.
The Jurkat cell line was established from the peripheral blood of a 14 year old boy by Schneider et al., and was originally designated JM.
The line was produced by treatment with ethylmethanesulfonate and negative selection with OKT3 monoclonal antibody.
This is a mutant line derived from the E6-1 clone of Jurkat (ATCC TIB 152) that lacks the beta chain of the T cell antigen receptor. |

| Comments: | xa0J.RT3-T3.5 cells lack the beta chain of the T cell antigen receptor. The cells do not express either CD3 or the T cell receptor alpha/beta heterodimer on the surface. The cells may be useful for transfection studies involving the T cell receptor beta chain gene. |
| Complete Growth Medium: | The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, ATCC 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (ATCC 30-2020) to a final concentration of 10%.

Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 1 x 105 cells/mL and maintain between 1 x 105 and 1 x 106 cells/mL. Do not allow the cell concentration to exceed 1xa0x 106 cells/mL. |
