Organism: Rattus norvegicus (B cell); Mus musculus (myeloma), rat (B cell); mouse (myeloma)
Cell Type: hybridoma: B lymphocyte
Product Format: frozen
Morphology: lymphoblast
Culture Properties: suspension
Biosafety Level: 1

Biosafety classification is based on U.S. Public Health Service Guidelines, it is the responsibility of the customer to ensure that their facilities comply with biosafety regulations for their own country.

Applications:
The antibody specifically binds to mouse ZP3 glycoprotein (amino acids 336-342).
The antibody cross-reacts with rat zonae but not with non-rodent species.
Derivation:
Spleen cells were fused with Sp2/0 mouse myeloma cells.
Genes Expressed:
immunoglobulin; monoclonal antibody; against mouse ZP3 glycoprotein
Cellular Products:
immunoglobulin; monoclonal antibody; against mouse ZP3 glycoprotein
Comments:
Animals were immunized with particulate murine zonae pellucidae.
Spleen cells were fused with Sp2/0 mouse myeloma cells.
During development and maturation, mammalian oocytes are surrounded by the zona pellucida, which in the mouse is comprised of three sulfated glycoproteins, ZP1, ZP2 and ZP3.
The zona pellucida mediates sperm binding at fertilization, provides a post-fertilization block to polyspermy, and facilitates passage of pre-implantation embryos down the oviduct.
The antibody specifically binds to mouse ZP3 glycoprotein (amino acids 336-342).
The antibody cross-reacts with rat zonae but not with non-rodent species.
Following passive immunization, this antibody completely blocks both in vivo and in vitro fertilization of subsequently ovulated mouse eggs.
Complete Growth Medium: Dulbeccos modified Eagles medium with 4 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate and 4.5 g/L glucose supplemented with 10 mM HEPES, 0.15 mg/ml oxaloacetate, 0.05 mg/ml pyruvate, 0.0082 mg/ml bovine insulin and 0.05 mM 2-mercaptoethanol, 80%; fetal bovine serum, 20%
Subculturing:
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)
Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 to 2 X 10 exp5 viable cells/ml.
Maintain cultures at cell concentrations between 5 X 10 exp4 and 5 X 10 exp5 viable cells/ml.
Attached cells may be subcultured by scraping.
Cryopreservation:
culture medium 95%; DMSO, 5%
Culture Conditions:
Temperature: 37.0°C
Isotype: IgG2a
Name of Depositor: J Dean
Deposited As: rat (B cell); mouse (myeloma)
Passage History:
The zona pellucida mediates sperm binding at fertilization, provides a post-fertilization block to polyspermy, and facilitates passage of pre-implantation embryos down the oviduct.
